摘要

Abstract

Objective To investigate the mechanism of action of Minor Xuming Decoction(小续命汤,MXMD)in astrocyte activation af-ter oxygen-glucose deprivation(OGD)/reoxygenation.Methods An in vitro model of OGD for 2.5 hours followed by reoxygenation for 24 hours was established to simulate cerebral ischemia-reperfusion injury.Human astrocyte U251 cells were randomly divided into the control group,the OGDn/reoxygenation group,the fluorocitrate group(0.25 mmol·L-1)as an astrocyte activation inhibitor,and a MXMD group(50 mg·L-1).Cell morphology was observed under an inverted microscope,LDH activity was detected by the dinitrophe-nylhydrazine method,ultrastructural changes were observed by transmission electron microscopy,and the fluorescence intensity of astro-cyte activation markers GFAP and S100βwas detected by immunofluorescence staining.Western blot(WB)was used to detect the ex-pression levels of GFAP and S100βproteins in each group.Results Compared with the control group,cells in the model group showed disintegration of the cell body,mitochondrial swelling or vacuolar changes,significantly decreasing cell viability,increasing LDH activity(P<0.05),and significantly increasing fluorescence intensity and protein expression levels of GFAP and S100β(P<0.05).Com-pared with the OGD/reoxygenation group,the fluorocitrate group and the MXMD group could improve cell morphology and ultrastructure,increase survival rate(P<0.05),reduce cell leakage rate(P<0.05),and significantly decrease the fluorescence intensity and pro-tein expression levels of GFAP and S100β(P<0.05).Conclusion MXMD may alleviate the injury induced by OGD/reoxygenation by inhibiting astrocyte activation,which may be related to the reduction of the protein expression levels of astrocyte activation markers GFAP and S100.
