芩百清肺浓缩丸抑制肺炎支原体感染小鼠炎症反应的作用机制研究
Study on the mechanism of Concentrated Qinbai Qingfei Pill(芩百清肺浓缩丸)in inhibiting the inflammatory response of mice with mycoplasma pneumoniae

Abstract
Objective To explore the mechanism of action of Concentrated Qinbai Qingfei Pill(百清肺浓缩丸,CQQP)in the treat-ment of mycoplasma pneumonia(MP).Methods Biacore binding technology was used to combine CQQP with AQP1 to analyze its bind-ing ability.Pathological sections of the mice lung tissues were prepared by animal experiments,and hematoxylin-eosin(HE)staining was performed to observe their pathological conditions.Immunohistochemistry(IHC)and RT-PCR were used to detect the effects of QQCP on the expressions of AQP1,JAK1 and STAT3 proteins and mRNA in the lung tissues.AQP1 and STAT3 were co-precipitated by co-immunoprecipitation technology,and the CO-IP results were visualized and analyzed by SDS-PAGE electrophoresis.Results The results of Biacore binding experiment showed that there was specific binding between CQQP and AQP1.The results of ani-mal experiments showed that CQQP significantly reduced the pathological damage of the lung tissues in MP-infected mice,up-regula-ted AQP1 protein and mRNA expression(P<0.05、P<0.01),and down-regulated JAK1 and STAT3 protein and mRNA expression(P<0.05、P<0.01).The results of CO-IP experiments showed that there was an interaction between AQP1 and STAT3.Conclu-sion CQQP treats MP by up-regulating AQP1 and down-regulating the expressions of JAK1 and STAT3 and inhibiting inflammatory re-sponse.关键词
