基于线粒体凋亡途径探讨化浊解毒方对CRF大鼠的作用

Abstract
Objective To explore the mechanism of Huazhuo Jiedu Formula(化浊解毒方,HZJDF)/Turbidity-resolving and Toxins-removing Formula on 5/6 nephrectomy rats based on the mitochondrial apoptosis pathway.Methods The successfully modeled SD rats were divided into the model group,the HZJDF low-dose,medium-dose and high-dose groups,the losartan potassium group,with 9 rats in each group.The liver and kidney function of CRF rats was detected by biochemical method.The pathological changes of renal tis-sues were observed by HE staining and Masson staining.The ultrastructural changes of mitochondria in renal tubular epithelial cells were observed by transmission electron microscopy(TEM).The apoptosis rate of renal tissue cells was detected by Tunel staining.The protein and mRNA expression levels of Bel-2,Bax,Cyt C,Caspase-3 and Caspase-9 were detected by Western blot(WB)and Real-time PCR.Results Compared with the sham operation group,the values of Cr and BUN were significantly increased(P<0.01).The me-sangial cells and stroma of the glomerulus were proliferating,the structure of the renal tubule interstitial area was disordered,a large number of blue collagen fibers were accumulated in the peribulbar and interstitial area,and the apoptosis rate of renal tissue cells was sig-nificantly increased(P<0.01).Renal tubular epithelial cell mitochondria were swollen,ridge was fractured,vacuolar degeneration happened.The expression levels of Bel-2 protein and mRNA were significantly decreased(P<0.05,P<0.01),and the expression levels of Bax,Cyt C,Caspase-3,Caspase-9 protein and mRNA were significantly increased(P<0.05,P<0.01).After the inter-vention of HZJDF,the values of Cr and BUN were decreased significantly(P<0.01).The above pathological damage was significantly relieved,the apoptosis rate of renal tissue cells decreased significantly(P<0.01).The expression levels of Bcl-2 protein and mRNA were significantly increased(P<0.05,P<0.01),and the expression levels of Bax,Cyt C,Caspase-3,Caspase-9 protein and mR-NA were significantly decreased(P<0.05,P<0.01).Conclusion HZJDF can inhibit mitochondrial apoptosis by regulating Bcl-2/Bax pathway.关键词
