玉屏风散对博来霉素诱导的肺纤维化大鼠中JAK2/STAT3信号通路的干预机制研究
Intervention mechanism of Yupingfeng Powder(玉屏风散)on JAK2/STAT3 Signaling pathway in rats with bleomycin-induced pulmonary fibrosis

Abstract
Objective To observe the regulatory mechanism of Yupingfeng Powder(玉屏风散,YPFP)on the pulmonary fibrosis activa-tion of the JAK2/STAT3 signaling pathway,and to explore the mechanism of YPFP in ameliorating idiopathic pulmonary fibrosis(IPF).Methods 48 SD rats were randomly divided into 8 groups,with 6 rats in each:the blank group,sham-operated group,model group,positive control group,STAT3 inhibitor group,and high/medium/low-dose YPFP groups,in which rats in the model group and in YPFP groups were injected with BLM intratracheally to establish the lung fibrosis model.Gastric administration was started after the 14th day of modeling,and prednisone(6 mg·kg-1·d-1)was given to the positive control group,sorafenib(30 mg·kg-1·d-1)was giv-en to the STAT3 inhibitor group,and YPFP decoction was given to the high-dose(6 g·kg-1·d-1),medium-dose(3 g·kg-1·d-1),and low-dose(1.5 g·kg-1·d-1)YPFP groups,respectively.Equal volume of normal saline was given to the blank group and model group.Drug was continuously administered until the 30th day.Symptoms and signs,changes in lung tissue mor-phology,changes in α-SMA protein expression,IL-6,TGF-β1 content,JAK2/STAT3 signaling pathway levels in lung tissues were observed in the respective groups before and after treatment.Results Compared with the blank group and the sham-operated group,rats in the model group showed severe symptoms,signs and lung tissue injury,significantly higher α-SMA protein expression and IL-6 and TGF-β1 content(P<0.01),and significantly higher JAK2,p-JAK2,STAT3 and p-STAT3 expressions(P<0.01).After drug treatment,compared with the model group,rats in the high-dose YPFP group,the prednisone group and the sorafenib group showed a significant alleviation in symptoms,signs and lung tissue damage,with a significant decrease in α-SMA protein expression and IL-6 and TGF-β1 contents(P<0.01),and a significant decrease in the expressions of JAK2,p-JAK2,STAT3 and p-STAT3(P<0.01).The medium-dose YPFP group showed a certain degree of improvement in the indexes(P<0.01 or P<0.05),while the inde-xes in the low-dose YPFP group basically showed no significant changes.Overall,the efficacy of the high-dose YPFP group was better than that of the medium-dose and low-dose groups,and was close to that of the prednisone group,with a better tendency than that of the sorafenib group.Conclusion YPFP is able to inhibit the phosphorylation and activation of the JAK2/STAT3 pathway by lowering the levels of IL-6 and TGF-β1,thus reducing the differentiation of myofibroblasts,lowering the expression of α-SMA,and decreasing the deposition of collagen fibers in the rat lung tissues,and ultimately achieving the purpose of ameliorating and treating IPF.关键词
