黑背胡狼源蜱的分子鉴定及pcox1、pnad5和ITS-1基因的遗传多样性分析
Molecular Identification of Ticks in Black-Backed Jackals and Genetic Diversity Analysis of pcox1、pnad5 and ITS-1 Genes

Abstract
To identify tick species parasitizing black-backed jackals(Canis mesomelas)and analyze their genetic diversity,six ticks were collected from the body surface of black-backed jackals at Changsha Ecological Zoo.Mitochondrial part of cytochrome c oxidase subunit 1(pcox1),part of nicotinamide adenine dinucleotide dehydrogenase subunit 5(pnad5),and internal transcribed spacer 1(ITS-1)genes were amplified via PCR and sequenced.Bioinformatics tools were used to assess sequence homology,haplotype diversity,nucleotide diversity,and to construct phylogenetic trees.The lengths of pcox1,pnad5,and ITS-1 sequences were 780,510,and 1 500 bp,respectively.Sequence homology compared with GenBank entries ranged from 99.9%-100%(pcox1),99.4%-100%(pnad5),and 98.2%-100%(ITS-1).Two,three,and four haplotypes were detected in pcox1,pnad5,and ITS-1 sequences,with haplotype diversities of 0.333±0.046,0.733±0.024,and 0.800±0.029 63,respectively,and nucleotide diversities of 0.000 43±0.000 000 3,0.001±0.000 001 7,and 0.007 78±0.000 003 8.Phylogenetic analysis showed that all six ticks clustered within the Haemaphysalis longicornis clade,closely related to known genus Haemaphysalis sequences,and more distantly related to other genera such as Argas persicus and Ornithodoros tartakovskyi.These results confirm that the ticks from black-backed jackals are H.longicornis,and the pcox1,pnad5,and ITS-1 genes are reliable molecular markers for species identification.This study provides reference data for the taxonomy of H.longicornis and prevention of tick-borne diseases in black-backed jackals.关键词
