摘要

Abstract

Objective To establish the quality standards of different Salviae Miltiorrhizae Radix Et Rhizoma extracts,and to investigate the effects of each extract on inflammatory factors in diabetic angiopathy model rats.Methods Qualitative identification of tanshinone ⅡA and salvianolic acid B in different solvent(water,50%ethanol,95%ethanol)extracts were performed by thin layer chromatography(TLC).The content of salvianolic acid B in the extracts were determined by high performance liquid chromatography(HPLC).The chromatography column was Diamonsil TM-C18 column(250 mm×4.6 mm,5 µm),the mobile phase was methanol-acetonitrile-formic acid-water(30:10:1:59,V/V/V/V),the flow rate was 1.0 mL/min,the detection wavelength was 286 nm,the column temperature was 30 ℃,and the injection volume was 10 µL.A total of 50 rats were randomly divided into the control group,the model group,the water extract group,the 95%ethanol extract group and the 50%ethanol extract group,with 10 rats in each group.Diabetic model rats were replicated by intraperitoneal injection of 1%streptozotocin(60 mg/kg)after one month of high-fat and high-sugar diet.After successful modeling,the corresponding extract(10 mL/kg)or normal saline was intragastrically administered once a day for two months.The fasting blood glucose(FBG)level of rats was measured by blood glucose test paper.The levels of serum interleukin(IL)-6,C-reactive protein(CRP),intercellular adhesion molecule(ICAM)-1 and cluster of differentiation 40 ligand(CD40L)were detected by enzyme-linked immunosorbent assay(ELISA).The expression of tumor necrosis factor(TNF)-α in rats was analyzed by immunohistochemistry.Results The TLC spots of tanshinone ⅡA and salvianolic acid B in 50%ethanol and 95%ethanol extracts were clear and well separated,and tanshinone ⅡA was not detected in the water extract.The linear range of salvianolic acid B was 0.525-2.625 µg(r=0.999 7,n=5).The RSDs of precision,stability and repeatability test results were all lower than 2.4%.The average recovery was 98.39%,with RSD of 1.28%(n=6).The content of salvianolic acid B in the samples was 50%ethanol extract(85.751 mg/g)>water extract(61.733 mg/g)>90%ethanol extract(43.335 mg/g).Compared with the model group,the levels of FBG,ICAM-1,CRP,CD40L,IL-6 in serum and TNF-α in abdominal aortic tissue were significantly decreased in the water extract group,95%ethanol extract group and 50%ethanol extract group(P<0.05).Conclusion The establishment of quality standards for different extracts is simple and the results are stable and reliable.The fat-soluble and water-soluble extracts in Salviae Miltiorrhizae Radix Et Rhizoma can reduce the inflammatory response of diabetic model rats.The mechanism may be related to the reduction of inflammatory factors by tanshinone ⅡA and salvianolic acid B.Among them,50%ethanol extract demonstrates the best hypoglycemic and anti-inflammatory effects.
