全真一气汤-艾灸序贯疗法通过自噬效应蛋白苄氯素1/同源性磷酸酶张力蛋白诱导激酶1/缺氧诱导因子-1α/水通道蛋白5信号通路改善慢性阻塞性肺疾病阳虚水泛证水代谢障碍机制研究

Abstract
Objective:This study aims to explore the mechanism of the Quanzhen Yiqi decoction(全 真 一 气 汤)plus moxibustion sequential therapy on water metabolism disorder of chronic obstructive pulmonary disease(COPD)with the Yang(阳)deficiency and water flood syndrome through BECN1/PINK1/HIF-1α/AQP5 signaling pathway.Methods:Fifteen male C57BL/6 mice were divided into three groups of five each:the control group,the model group,and the Chinese medicine+moxibustion group.COPD models in mice with the Yang deficiency and water flood syndrome were established based on previous studies.Changes in renal function and renal tissue were observed via cystatin levels,transmission electron microscopy,and histopathological staining after administration of the Quanzhen Yiqi decoction plus moxibustion sequential therapy.Western blot was used to detect the expression of BECN1,PINK1,HIF-1α and AQP5 proteins in renal tissues.Results:Compared with the control group,the model group showed a significant increase in serum Cys-C levels(P<0.05).HE staining revealed enlarged kidney volume,mild glomerular atrophy,and plasma protein extravasation in the model group.Electron microscopy showed significant cellular damage in the model group,including organelle condensation,abundant vacuoles,mitochondrial structural damage,and autophagic vacuoles.Western blot analysis demonstrated a significant increase in HIF-1α levels and significant decreases in AQP5,BECN1,and PINK1 levels in the model group(P<0.05).Compared with the model group,the Chinese medicine+moxibustion group showed a significant decrease in serum Cys-C level after intervention(P<0.05).HE staining showed reduced kidney volume and no obvious glomerular atrophy after Chinese medicine+moxibustion intervention.Electron microscopy revealed no significant cellular damage,vacuoles,or autophagic vacuoles after combined intervention.Western blot analysis showed a significant decrease in HIF-1α levels and significant increases in AQP5,BECN1,and PINK1 levels in renal tissues after Chinese medicine+moxibustion intervention(P<0.05).Conclusion:The sequential therapy of the Quanzhen Yiqi decoction plus moxibustion regulates the"lung-kidney axis"through the BECN1/PINK1/HIF-1α/AQP5 signaling pathway,ameliorating water metabolic disorders in COPD with the Yang deficiency and water flood syndrome.关键词
