A study on the pharmacological substance basis and action mechanism of Huashanshen Diwan in the treatment of COPD based on UPLC-Q-TOF-MS/MS technology and network pharmacology

Abstract
Objective:To explore the pharmacological substance and underlying action mechanism of Huashanshen Diwan(华 山参滴丸)in the treatment of chronic obstructive pulmonary disease(COPD).Methods:The components in plasma of Huashanshen Diwan were analyzed by UPLC-Q-TOF-MS/MS technology,and the active ingredients were screened out by the Swiss ADME platform further.The active ingredient corresponding targets were searched by the Swiss Target Prediction and SuperPred databases,and COPD disease targets were sorted out by the GeneCards database.The intersection targets of ingredient targets and disease targets were taken to obtain underlying targets of Huashanshen Diwan in the treatment of COPD.A protein-protein interaction network was constructed by STRING,and a medicine-active ingredient-intersection target interaction network was obtained by Cytoscape 3.9.1 software to further screen out core targets.The GO functional enrichment analysis and KEGG pathway enrichment analysis were performed to explore the possible action mechanism of underlying targets,and molecular docking validation was performed on core active components and core targets by CB-Dock.Results:A total of 19 components in plasma were identified from Huashanshen Diwan,and 288 underlying targets were screened out,of which 56 were core targets.The GO functional enrichment analysis and KEGG pathway enrichment analysis showed that calcium signaling pathway,PI3K/Akt signaling pathway,cAMP signaling pathway,MAPK signaling pathway,Ras signaling pathway were the key signaling pathways of Huashanshen Diwan in the treatment of COPD,and ESR1,ALB,EGFR,STAT3,and PTGS2 were the key targets.The results of molecular docking showed that formononetin,biochanin A,anisodamine,scoparone,palmic acid had good binding activity with the core targets,which were the main active substances of Huashanshen Diwan in the treatment of COPD.Conclusion:Huashanshen Diwan can treat COPD with multiple components,multiple targets,and multiple pathways,providing a basis for in-depth biological experiment.关键词
