Abstract
Objective:To explore the mechanism of Jiawei Shoutai Wan(加味寿胎丸)in improving endometrial receptivity using network pharmacology and molecular docking technology.Methods:Active ingredients of Jiawei Shoutai Wan were screened by TCMSP.Disease-related targets were identified through the GeneCards database,and a Venn diagram was used to obtain intersecting targets.The STRING database was used to construct a protein-protein interaction network,and Cytoscape was employed to visualize the active ingre-dient-target-disease network.The DAVID database was utilized for GO functional enrichment analysis and KEGG pathway enrichment analysis.Molecular docking verification was performed on core ingredients and potential targets using AutoDock and PyMOL software.Results:Through comprehensive database analysis,28 active ingredients were identified,with 90 corresponding targets for the effective ingredients and 922 targets related to endometrial receptivity.Network topology analysis identified quercetin,kaempferol,β-sitosterol as key active ingredients,with core targets mainly involving TNF,IL-6,and AKT1.GO analysis revealed that the active ingredients of Jiawei Shoutai Wan mainly participated in the positive regulation of gene expression,inflammatory response,cellular response to stimuli,and positive regulation of endothelial cell proliferation.KEGG analysis uncovered regulatory mechanisms,involving key pathways such as AGE-RAGE,IL-17,Toll-like receptor,HIF-1,estrogen,and TNF.Molecular docking results showed that the core ingredients exhibited strong binding affinity with key targets,with predicted binding sites and interaction patterns demonstrating good stability.Conclusion:The active ingredients of Jiawei Shoutai Wan may improve endometrial receptivity by targeting key molecules such as TNF and IL-6,activating core signaling pathways such as AGE-RAGE and IL-17,regulating moderate proliferation and apoptosis in the endometrium,and promoting angiogenesis,anti-inflammatory,and immue regulation effects,thus playing a role in improving endometrial receptivity.关键词
