基于网络药理学及分子对接探析黄柏-知母治疗儿童特发性性早熟的潜在作用机制
The potential action mechanism of Huangbo-Zhimu in the treatment of idiopathic precocious puberty in children based on network pharmacology and molecular docking

Abstract
Objective:To explore the potential action mechanism of Huangbo(Cortex Phellodendri Chinsis)-Zhimu(Rhizoma Anemarrhenae)in the treatment of idiopathic precocious puberty in children by network pharmacology and molecular docking.Methods:Clinical randomized controlled trials of Chinese medicine treatment for idiopathic precocious puberty in children were collected from CNKI,CBM,Wanfang,and VIP.The used frequency and association rules of relevant Chinese medicines were analyzed to screen out core medicines.The TCMSP was used to screen out the active ingredients of core medicines.The Human Phenotype Ontology database,DisGeNET database,and GeneCards database were used to obtain potential disease-related targets.The intersection of medicinal active ingredients and disease targets was gained for protein-protein interaction analysis.The DAVID was used to perform GO functional enrichment analysis and KEGG pathway enrichment analysis on relevant targets to explore their potential biological functions and signaling pathways.Molecular docking was used to predict the binding ability of active ingredients with target proteins.Results:The core medicines for treating idiopathic precocious puberty in children were Huangbo and Zhimu.A total of 217 potential active ingredients and 212 potential targets were obtained.The main active ingredients of the core medicines were quercetin,kaempferol,icariin,graveoline,and stigmasterol.The key targets were TP53,AKT1,EGFR,ESR1,IL-6,and TNF,and the main signaling pathways involved were MAPK signaling pathway,PI3K-Akt signaling pathway,and estrogen signaling pathway.The molecular docking result showed that the main active ingredients and core targets were well combined.Conclusions:Huangbo-Zhimu mainly regulate MAPK,PI3K-Akt,and estrogen signaling pathways by acting on key targets such as TP53,AKT1,EGFR,ESR1,IL-6,and TNF,thus affecting endocrine metabolism and exerting therapeutic effects on idiopathic precocious puberty in children.Precocious puberty may affect glucose and lipid metabolism and cardiac function of children patients.关键词
