The action mechanism of Shengjiang San in the treatment of non-alcoholic fatty liver disease based on network pharmacology and molecular docking

Abstract

Objective:Through network pharmacology and molecular docking technology,the action mechanism of Shengjiang San(升降散)in the treatment of non-alcoholic fatty liver disease(NAFLD)was explored preliminarily.Methods:Through TCMSP and Swiss Target Prediction,active ingredients and action targets of Shengjiang San in exerting pharmacological effects were obtained.The GeneCards,OMIM,and TTD databases were used to obtain NAFLD related targets.The interaction analysis of the active ingredient targets of Shengjiang San and targets related to NAFLD was performed to find potential therapeutic targets of Shengjiang San in treating NAFLD.Cytoscape 3.7.2 software was used to construct a network map of TCM compounds-Chinese medicines-targets,and protein-protein interaction network analysis was performed by STRING database.GO functional analysis and KEGG pathway enrichment analysis were performed on key targets by DAVID database to screen out key action targets and main active components.Finally,the molecular docking verification was performed by AutoDockTools 1.5.6 software.Results:A total of 29 active ingredients in Shengjiang San,1 318 NAFLD related targets,and 88 potential therapeutic targets were predicted,and 140 KEGG pathways were obtained.Among the above enrichment pathways,PI3K/Akt signaling pathway,MAPK signal pathway,and AGE-RAGE signaling pathway might play an important role in the treatment of NAFLD with Shengjiang San.Conclusion:Shengjiang San may treat NAFLD through multiple targets and multiple pathways,among which AKT1,TNF,IL-1β,TP53,and ESR1 are the key target proteins of Shengjiang San in intervening in NAFLD.Shengjiang San may be involved in regulating PI3K/Akt,MAPK,and AGE-RAGE and other signaling pathways through the above key proteins to regulate cell proliferation and apoptosis,inhibit inflammatory responses and excessive fibrosis,thus playing a role of protecting liver.
