基于网络药理学和分子对接探讨补中益气汤治疗膈肌功能障碍的作用机制
The action mechanism of the Buzhong Yiqi decoction in the treatment of diaphragmatic dysfunction based on network pharmacology and molecular docking

Abstract
Objective:To explore the action mechanism of the Buzhong Yiqi decoction(补中益气汤)in the treatment of diaphragmatic dysfunction through network pharmacology and molecular docking.Methods:The active ingredients and targets of the Buzhong Yiqi decoction were retrieved through the TCMSP database.The disease-related targets were screened by GeneCards,OMIM,and NCBI databases,and the venn diagram was drawn to obtain the intersection targets of medicine and disease of the Buzhong Yiqi decoction on diaphragmatic dysfunction.The target network diagram was constructed by Cytoscape to screen out active ingredients.The protein-protein interaction network diagram of the medicine-disease intersection targets was constructed,and the core targets were obtained by cytohubba plug-in analysis.The GO analysis,KEGG analysis,and Reactome enrichment analysis were performed.Molecular docking was used to observe the binding ability of the active ingredients of the Buzhong Yiqi decoction to the core targets.Results:There were 163 active ingredients and 204 targets of the Buzhong Yiqi decoction,and 60 targets intersected with diaphragmatic dysfunction.The top five active ingredients were isorhamnetin,kaempferol,quercetin,β-sitosterol,and stigmasterol,respectively.The top five core genes were HIF-1α,BcL2,EGFR,MMP9,and PTGS2.Enrichment analyses showed that the Buzhong Yiqi decoction mainly regulated IL-17,HIF-1,and EGFR signaling pathways,involved the regulation of cell apoptosis,cytokine activity,immune response and other processes.Molecular docking results showed that the active ingredients of the Buzhong Yiqi decoction had good binding activity with the core targets.Conclusion:The Buzhong Yiqi decoction may act on HIF-1α,BcL-2,MMP9 and other core targets through active ingredients such as isorhamnetin and kaempferol to regulate HIF-1 and other signaling pathways,exerting synergistic effects of anti-oxidative stress,anti-inflammation,anti-apoptosis,anti-fibrosis,and tissue repair,so as to treat diaphragmatic dysfunction.关键词
