基于网络药理学探讨柯孜木克颗粒治疗慢性细菌性前列腺炎作用机制
Mechanism of Kezmekh granules in the treatment of chronic bacterial prostatitis based on network pharmacology

Abstract
Based on network pharmacology and animal experiments,this study investigated the preventive and thera-peutic effects of Kezmekh(KZMK)granules,a traditional Chinese medicine and ethnic medicine,on chronic bacterial prostatitis(CBP).Network pharmacology methods were used to predict the core targets and signaling pathways of KZMK granules acting on CBP,and a total of 181 potential targets were screened,primarily involving four key signaling pathways,with KEGG enrichment analysis confirming the phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT)sig-naling pathway as the most significantly associated with CBP.To validate these predictions,the study successfully estab-lished a rat CBP model induced by Escherichia coli.Hematoxylin and eosin(HE)staining revealed that KZMK signifi-cantly improved prostate tissue histopathology and reduced inflammatory cell infiltration.Enzyme-linked immunosorbent assay(ELISA)demonstrated that KZMK markedly decreased serum levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)(P<0.05 or P<0.01).Western blot analysis confirmed that KZMK significantly downregulated the expression of PI3K,AKT,as well as TNF-α,IL-1β,and IL-6 proteins in prostate tissues(P<0.001 or P<0.01).In summary,these findings suggest that KZMK may exert its therapeutic effects on CBP by inhibiting the re-lease of pro-inflammatory factors IL-6 and TNF-α,regulating the phosphorylation of key proteins in the PI3K-AKT sig-naling pathway,thereby alleviating inflammatory responses in prostate tissues.关键词
