藏绵羊睾丸组织VIM表达量的定量研究
Quantitative study on the expression of VIM in testicular tissues of Tibetan sheep

Abstract
This study aimed to investigate the differential expression of the vimentin(VIM)gene in the testicular tissues of Tibetan rams with different genotypes,and to further analyze the relationship between VIM expression and the reproductive performance of Tibetan rams,providing a theoretical basis for identifying molecular markers related to ram fertility.Six healthy Tibetan rams were selected and divided into B+type and++type according to their genotypes.Testicular tissue samples were collected,total RNA was extracted and reverse transcribed into cDNA.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the expression level of VIM gene in testicular tissues of different genotypes of Tibetan rams.At the same time,the expression level of VIM protein in different genotype tissues was detected by Western blot as verification.The results showed that the expression of VIM genetype in the testis of++type Tibetan rams was significantly higher than that of B+type(P<0.05),and the relative expression of protein was also higher than that of B+type,suggesting that the expression of VIM gene was closely related to the fertility of Tibetan rams,indicating that VIM may play an important role in the development of testis and the regulation of reproductive function.关键词
