体外生产绵羊早期胚胎发育异常分析
Analysis of developmental abnormalities in early sheep embryos produced in vitro

Abstract
Early developmental abnormalities of embryos in domestic animals represents a considerable economic loss.The experiment aimed to investigate the early developmental abnormalities of in vitro produced sheep embryos.Immature oocytes from sheep and mice were collected and used for in vitro maturation and fertilization,respectively.During the in vitro embryo culture,the developmental processes were tracked for counting the normal developmental rates and the proportions of embryos at different stages of development at different time points.Subsequently,immunofluorescence staining was used to detect the cell proliferation and differentiation of sheep blastocysts.The results showed that the blastocyst formation and hatching rates of in vitro produced sheep embryos were significantly lower compared to those of either in vitro produced mouse embryos or sheep embryos(P<0.05).About 11.27%and 53.90%of in vitro produced sheep embryos were arrested at the 2~4 cell stage and 8~16 cell stage,respectively.The developmental delay was observed in the blastulation of some sheep embryos.Moreover,despite more trophectoderm cells in sheep blastocysts,the proportion and proliferation activity of trophectoderm cells were lower than in mouse blastocysts.In conclusion,the in vitro produced sheep embryos showed a high incidence of developmental abnormalities,primarily characterized by the arrest at the 8~16 cell stage and the delayed blastocyst development.关键词
