早胜牛mtDNA D-loop区遗传多样性及起源分析
Analysis on Genetic Diversity and Origin of the mtDNA D-loop Region of Zaosheng Cattle

Abstract
To effectively protect the genetic resources of the Zaosheng cattle and enhance their produc-tion traits while mitigating the negative effects of mutation,genetic drift,and inbreeding,this study em-ployed PCR amplification,sequencing,and bioinformatics to investigate the sequence polymorphism and maternal lineage by targeting the mitochondrial DNA(mtDNA)control region(D-loop region)in Zao-sheng cattle.The results showed that a total of 84 nucleotide sequences of approximately 910 bp and one se-quence of 953 bp were successfully obtained from 98 samples of Zaosheng cattle.Analysis revealed that ade-nine(A)had the highest average proportion,32.92%,followed by Thymine(T,28.75%.),cytosine(C,24.57%),and guanine(G,13.71%),indicating a high consistency in the length of the D-loop region across individuals.Further analysis showed that there were 60 effective polymorphic sites were identified within the mtDNA D-loop,resulting in 45 distinct haplotypes and a high haplotype diversity index of 0.946±0.014.The nucleotide diversity was 0.017 84±0.002 15,reflecting a moderate degree of sequence variabili-ty,neither highly conserved nor overly divergent.The genetic distances between the 45 haplotypes ranged from 0.013 to 1.508,with an average of 0.088.The minimum genetic distance of 0.013 indicated that some haplotypes might have originated from a common ancestor and had a relatively recent divergence time.The results of the phylogenetic tree analysis revealed that the 45 haplotypes of Zaosheng cattle were closely related to Bos taurus and Bos indicus,especially most of the haplotypes(such as Hap 17,Hap 39,Hap 43,etc.)were more closely aligned with Bos taurus.These findings suggest that Bos taurus likely represents the predominant maternal origin of Zaosheng cattle.关键词
