Synthesis and Anti-Staphylococcus Aureus Activity of N,N'-Dioctylimidazolium-Based N-Heterocyclic Carbene Silver/Gold Complexes

摘要

Abstract

Objective:N-heterocyclic carbene(NHC)-gold(I)(C8Au)and NHC-silver(I)(C8Ag)complexes were synthesized using 1,3-dioctylimidazolium ionic liquid as a precur-sor.Their antibacterial activities were evaluated,particularly against Staphylococcus au-reus.Methods:The structures of the complexes were characterized by 1H-NMR and HRMS.Their antimicrobial activity and safety were assessed through minimum inhibitory concentration(MIC),time-kill assay,biofilm inhibition and eradication assays,drug re-sistance tests,checkerboard synergy assays,and a Galleria mellonella larvae model.Re-sults:C8Ag demonstrated significantly superior antibacterial activity against Staphylococcus aureus compared to C8Au,achieving rapid bactericidal effects within 2 hours.C8Ag also ex-hibited excellent biofilm inhibition and eradication capabilities,with a low tendency to induce drug resistance.Additionally,C8Ag showed strong synergistic effects when combined with various antibiotics.The Galleria mellonella larvae model confirmed the significant in vivo antibacterial efficacy and safety of C8Ag.Conclusion:The octyl chain-modified C8Ag com-plex exhibits significant antibacterial effects both in vitro and in vivo,demonstrating promis-ing potential for future applications.
