摘要

Abstract

Objective:To explore the effects of artesunate(ART)on the biological behavior of human oral squamous cell carcinoma cells and its mechanism of action.Methods:HSC-4 cells were treated with different concentrations(0,6.25,12.5,25,50 μg/mL)of ART.Cell viability was detected by CCK-8 assay.The number of invasive cells was detected by Tran-swell chamber assay.Cell migration rate was detected by scratch assay.The clone formation rate was detected by plate clone assay.The expression levels of heme oxygenase-1(HMOX-1)in each group were detected by RT-qPCR and Western blot.HMOX-1 interference adenovirus vector(si-HMOX-1)and its control(si-NC)were transfected into HSC-4 cells,and then trea-ted with 50 μg/mL ART.The control(Con)group,ART group,ART+si-NC group and ART+si-HMOX-1 group were set up.The biological behaviors of cells in each group were detected by CCK-8,Transwell,scratch,plate clone,RT-qPCR and West-ern blot assays.The levels of reactive oxygen species(ROS)and apoptosis rate in each group were detected by flow cytometry.Parallel experiments were set up and the above detections were conducted using KOSC-2cl3-43 and Tca8113 cells.Results:The cell viability,number of invasive cells,migration rate and clone number of HSC-4 cells treated with ART decreased in a dose-dependent manner,while the expression levels of HMOX-1 mRNA and protein increased in a dose-dependent manner.Among them,the effect of 50 μg/mL ART concentration was the best.ART could significantly inhibit the proliferation,inva-sion and migration of HSC-4 cells,and significantly increase the ROS level and apoptosis rate(P<0.05).Down-regulation of HMOX-1 could significantly reverse the inhibitory effect of ART on the proliferation,invasion and migration ability of HSC-4 cells.ART inhibited the proliferation,invasion and migration of KOSC-2cl3-43 and Tca8113 cells by up-regulating the expres-sion of HMOX-1.Conclusion:ART inhibits the malignant biological behaviors such as proliferation,invasion and migration of human oral squamous cell carcinoma cells by up-regulating the expression of HMOX-1.
