摘要

Abstract

This study isolates and screens a fungal strain from tea leaves using a tannase identification medium.The strain is identified as Aspergillus niger through morphological characteristics and ITS sequence analysis,and designated as strain ALF11-1.Fermented with the spore suspension of strain ALF11-1,the tannase production activity of black tea leaf stalks reaches 10.42 U/mL.Subsequently,the optimal fermentation conditions are determined through one-factor and response surface experiments,which are as follows:temperature 31℃,incubation time 4 d,inoculum size 4%,and ammonium chloride concentration 7%.Under these conditions,the predicted enzyme activity is 24.9 U/mL,while the measured activity reaches 25.92 U/mL.Compared to the pre-optimization enzyme activity(10.4 U/mL),the optimized solid-state fermentation conditions enhance tannase production by strain ALF11-1 approximately 2.49-fold.
