Abstract

Human apurinic/apyrimidinic endonuclease 1(APE1),as a crucial enzyme within the base excision repair(BER)pathway,exhibits abnormal expression in tumor cells and serves as one of the biomarkers for cancer diagnosis.Therefore,the development of highly sensitive methods for detecting APE1 activity holds significant research value.This study,based on the high walking efficiency and stability of the bipedal DNA walker,utili-zes exonuclease Ⅲ(Exo Ⅲ)as the driving force and gold nanoparticles(AuNPs)as the carriers of the track strands.Leveraging the fluorescence quenching mechanism,a bipedal DNA walker fluorescence biosensor for the highly sensitive detection of APE1 has been developed.Under optimal detection conditions,the activity of APE1 exhibited a favorable linear relationship with the fluorescence intensity of the system within the range of 0.01 U/mL to 0.5 U/mL.The linear equation was determined to be F=173.00CAPE1+99.45,with a detection limit of 0.006 U/mL.This method can be applied to detect APE1 activity in serum-mimicking samples,with a spike re-covery rate ranging from 96.4%to 98.5%.
