Double-stranded DNA Cytosine Deaminase Detects DNA Methylation
摘要
Abstract
A highly sensitive double stranded DNA cytosine deaminase DddA coupling methylation-specific PCR method was developed for detecting DNA methylation,avoiding DNA sample degradation in bisulfite conversion coupled Methylation-Specific PCR.In Dd-dA conversion-based methylation specific real-time quantitative PCR,the DddA deamina-ted the DNA samples firstly,then performed the real-time PCR with methylation specific primers.According to standard curve of the target genes,the methylation level of the DNA sample could be evaluated.The results showed that the DddA can discriminate the methylate with unmethylated DNA well.And DddA-qMSP can effectively detect the methylation status of colorectal cancer marker genes SEPT9 and SDC2.关键词
