黑果枸杞花青素通过介导Nrf2和NF-κB通路缓解低氧诱导的H9c2心肌细胞氧化应激
Anthocyanin of Lycium barbarum attenuate hypoxia-induced oxidative stress in H9c2 cardiomyocytes via the Nrf2 and NF-κB signaling pathways

Abstract
To investigate the underlying molecular mechanisms of anthocyanin of Lycium barbarum(AC)on hypoxia-induced oxidative stress in H9c2 cardiomyocytes and their alleviating effects.A hypoxic H9c2 cardiomyocyte model was established under 1%O2,the mRNA and protein expression levels of key factors in the Nrf2 and NF-κB signaling pathways were analyzed using qRT-PCR and Western-blot under normoxic and hypoxic conditions following AC treatment.The results demonstrated that hypoxia signifi-cantly(P<0.05)reduced mRNA expression of Nrf2 pathway components(KAP1,Nrf2,HO-1,and NQO1),with Nrf2 and KAP1 protein levels showing extremely significant decreases(P<0.01),while HO-1 andNQO1 proteins were significantly downregulated(P<0.05).Compared to the hypoxia group,the hypoxia+AC group exhibited extremely significant upregulation of both mRNA and protein levels of KAP1,Nrf2,HO-1,and NQO1(P<0.01).Hypoxia induced extremely significant(P<0.01)increases in mRNA and protein expression of NF-κB pathway components(IKK-α,IKK-β,NF-κB,TNF-α,and COX-2).AC treatment under hypoxia significantly suppressed these increases(P<0.01).Further experiments using the NF-κB activator imiquimod revealed that the hypoxia+imiquimod group exhibited extremely significant upregulation of IKK-β and NF-κB proteins(P<0.01)and significant increases in IKK-α,TNF-α,and COX-2 proteins(P<0.05).In contrast,the hypoxia+AC+imiquimod group showed extremely significant downregulation of all these proteins(P<0.01).These findings indicate that AC alleviates hypoxia-induced oxidative stress in H9c2 cardiomyocytes by activating the Nrf2 pathway and suppressing the NF-κB signaling pathway,thereby providing cardiomyocyte protection.关键词
