甲胎蛋白调控HIPPO信号通路对肝细胞癌细胞重编程基因表达的影响OA
Effect of Alpha-fetoprotein Regulating the HIPPO Signaling Pathway on the Expression of Reprogramming Genes in Hepatocellular Carcinoma Cells
目的 探讨甲胎蛋白(AFP)调控HIPPO信号通路对肝细胞癌(HCC)细胞重编程基因表达的影响.方法 构建稳定下调AFP的HUH7 细胞系和稳定上调AFP的HLE细胞系,依据转染病毒的不同分为HUH7 组(不转染)、HUH7C组(转染LV-NC)、HUH7-siAFP组(转染LV-siAFP)、HLE组(不转染)、HLEC组(转染LV-NC)、HLE-AFP组(转染LV-AFP).通过western bloting检测AFP对重编程基因表达的影响,通过Discovery studio 软件模拟AFP与YAP的蛋白互作,GEPIA2在线数据库分析YAP与重编程基因的相关性,平板克隆检测下调或上调AFP对HCC细胞克隆形成的影响,免疫荧光实验观察下调或上调AFP对YAP蛋白在HCC细胞胞质和胞核分布的影响,CO-IP实验检测HUH7 细胞中AFP与YAP蛋白互作.结果 上调AFP可促进HCC细胞重编程基因的表达,下调AFP可抑制HCC细胞重编程基因的表达,GEPIA2 在线数据库分析YAP与重编程基因表达存在正相关,Discovery studio软件模拟、CO-IP实验和免疫荧光实验提示AFP和YAP存在互作,AFP可影响YAP的核定位.结论 在HCC细胞中,AFP可能通过促进HIPPO信号通路中的YAP在细胞核中的聚集从而增强其下游重编程基因的表达.
Objective To explore the effect of alpha-fetoprotein(AFP)regulating the HIPPO signaling pathway on the expression of reprogramming genes in hepatocellular carcinoma(HCC)cells.Methods Construct the HUH7 cell line with stable down-regulation of AFP and the HLE cell line with stable up-regulation of AFP According to the different transfected viruses,they were divided into the HUH7 group(untransfected),the HUH7C group(transfected with LV-NC),the HUH7-SIAFP group(transfected with LV-siAFP),HLE(untransfected),HLEC(transfected with LV-NC),and HLE-AFP(transfected with LV-AFP).The effect of AFP on the expression of reprogramming genes was detected by western bloting.The protein interaction between AFP and YAP was simulated by Discovery studio software.The correlation between YAP and reprogramming genes was analyzed by GEPIA2 online database.The effect of down-regulation or up-regulation of AFP on the colony formation of HCC cells was detected by plate cloning.The effect of down-regulation or up-regulation of AFP on the distribution of YAP protein in the cytoplasm and nucleus of HCC cells was observed by immunofluorescence assay.The interaction between AFP and YAP protein in HUH7 cells was detected by CO-IP assay.Results Upregulation of AFP can promote the expression of reprogramming genes in HCC cells,while downregulation of AFP can inhibit the expression of reprogramming genes in HCC cells.GEPIA2 online database analysis showed a positive correlation between YAP and reprogramming gene table.Discovery studio software simulation,CO-IP experiment,and immunofluorescence experiment suggested that AFP and YAP interact,and AFP can affect the nuclear localization of YAP.Conclusion In HCC cells,AFP may enhance the expression of downstream reprogramming genes by promoting the aggregation of YAP in the HIPPO signaling pathway in the nucleus.
