紫荆花粉萌发液体培养基的筛选及培养条件优化
Screening of Liquid Medium for in Vitro Germination and Optimization of Culture Conditions of Cercis chinensis Pollen

Abstract
In order to screen out the most suitable components of liquid culture medium of Cercis chinensis,the pollen of Cercis chinensis was used as the experimental material,and the liquid culture method was adopted to explore the effects of sucrose,boric acid,calcium nitrate,different culture temperatures and times on the germination of Cercis chinensis pollen.The results showed that the suitable medium components for pollen in vitro germination were:20%sucrose+200 mg/L boric acid+100 mg/L calcium nitrate.At this time,the pollen germination rate reached 86.43%,and the length of the pollen tube was 175.37μm.The appropriate in vitro culture time was 5 hours and the culture temperature was 25℃.Temperature was a key influencing factor.Unsuitable temperature could lead to a decrease in the activity of protective enzymes,disrupt the balance of reactive oxygen species me-tabolism,intensify membrane lipid peroxidation,and hinder pollen germination and pollen tube growth.关键词
