外泌体circPFKP诱导结直肠癌耐药性和致瘤性的研究
Study on Resistance and Tumorigenicity of Exosomal circPFKP in Inducing Colorectal Cancer

Abstract
Objective To explore the mechanism of exosomal circPFKP inducing 5-FU resistance and tumorigenicity in colorectal cancer(CRC)by targeting miR-665/LASP1.Methods High-throughput sequencing data of 50 pairs of CRC tissues and adjacent tissues were obtained from the GEO database GSE126094.Through quantification and background correction,the expression values of circRNAs in CRC tissues and adjacent normal tissues were obtained.The cell proliferation inhibition rate was determined by the MTT method.Exosomes were isolated from the culture medium of HCT116 and HCT116/5-FU cells.Online prediction software starBase,CircInteractome and circBank were used to predict the directly related miRNAs.Results It was found in qRT-PCR detection that the expression of circPFKP in CRC tissues was significantly up-regulated compared with adjacent normal tissues(P<0.05),and the expression of circPFKP in CRC cells was also significantly increased compared with normal cells(P<0.05).Knockout of circPFKP could increase the sensitivity of HCT116/5-FU to 5-FU(P<0.05).HCT116/5-FU/exo could significantly increase the IC50 concentration of HCT116 to 5-FU(P<0.05).CircPFKP could bind to miR-665(P<0.05),and LASP1 could bind to miR-665(P<0.05).Overexpression of miR-665 could inhibit the expression of LASP1(P<0.05).Conclusion CircPFKP may shuttle from drug-resistant CRC cells to adjacent sensitive CRC cells through exosomes,thereby inducing resistance to 5-FU.CircPFKP/miR-665/LASP1 is involved in regulating the growth mechanism of drug-resistant CRC cells.关键词
