Abstract

This study aimed to investigate the diversity and dynamics of endogenous inhibitors during the germination of Paris polyphylla var.Chinensis seeds.Employing seeds at various germination stages,systematic solvent extraction was utilized to isolate endogenous inhibitors,followed by their qualitative and quantitative analysis.This approach led to the identification of 17 compounds exhibiting inhibitory effects on seed germination.To further ascertain the pivotal endogenous inhibitors,cabbage,wheat,and mung bean seeds were employed as models to assess the half-maximal inhibitory concentrations(IC50)of hydroquinone.The IC50 values for hydroquinone were determined as 182.81,845.28,and 1510.08 mg/L,respectively.Additionally,the absolute content variation of hydroquinone were monitored throughout the germination course of Paris polyphylla var.Chinensis seeds.The results revealed a consistent decline in hydroquinone levels,with a pronounced decrease observed at a critical germination juncture(day 20).The findings illuminate the presence of multiple endogenous inhibitors within Paris polyphylla var.Chinensis seeds and single out hydroquinone as a crucial determinant of germination initiation.Notably,this work is the first to establish a direct correlation between decreased hydroquinone levels and the alleviation of seed dormancy in Paris polyphylla var.Chinensis.Collectively,this research not only provides a comprehensive profile of endogenous inhibitor dynamics during seed germination but also elucidates the pivotal role of hydroquinone in this process,thereby laying a robust foundation for the targeted development of dormancy-breaking techniques and enhancing the propagation efficiency of Paris polyphylla var.Chinensis seeds.
