基于网络药理学和实验验证探究肉苁蓉治疗阿尔茨海默症作用机制
Mechanism of Cistanche deserticola treating Alzheimer's disease based on network pharmacology and experimental validation

Abstract
Objective:To explore the potential action targets and optimal dosage of Cistanche deserticola on Alzheimer's dis-ease(AD)through network pharmacology and animal experiments.Methods:A total of 40 male 7-month-old SAMP8 mice were randomly divided into the model control group(Model),the low-dose(L-GCs),the medium-dose(M-GCs),and the high-dose(H-GCs)treatment group,and 10 male anti-aging SAMR1 mice of the same age were used as the SAMR1 control group.After 30 d of intragastric administration of GCs,the Morris water maze and Y maze experiments were used to test the learning and mem-ory abilities of each group of mice.Nissl staining was used to observe the number and morphology of neurons in the hippocampal CA1 region of the mice.Immunohistochemical staining and Western blot experiments were used to detect the expression of PI3K,P-Akt,and P-Tau in the hippocampus of mice.Results:A total of 8 effective components were screened from Cistanche desertico-la,and the core genes involved in AD treatment were Akt1,TNF,CASP3,etc.The signaling pathways related to AD treatment by Cistanche deserticola included PI3K-Akt,TNF,and AGE-RAGE pathways.Animal experiments showed that compared to the model group,the escape latency and total distance in the Morris water maze navigation test of the GC groups were significantly decreased(P<0.05),and the number of platform crossings were significantly increased(P<0.05)in the spatial exploration test of the GCs group.In the immunohistochemical staining and Western blot experiments,the ratio of PI3K and P-Akt/Akt in the GCs group were significantly increased(P<0.05),while the expression of P-Tau/Tau were significantly decreased(P<0.05).Conclusion:GCs may improve the learning and cognitive abilities of SAMP8 mice by regulating the PI3K-AkT signaling pathway and regulating the expression of P-Tau protein.关键词
