miR-508-3p inhibits proliferation and invasion of hepatocellular carcinoma cells by targeting CLEC1B gene

Abstract

Objective To explore the molecular mechanism of miR-508-3p regulating the proliferation and invasion of hepatocellular carcinoma cells.Methods Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)was used to detect the expres-sion levels of miR-508-3p and CLEC1B genes in 30 cases of liver cancer with definite pathological diagnosis and corresponding adja-cent normal tissues.The expressions of miR-508-3p and CLEC1B in human normal liver cells LO2 and hepatoma cells Huh7,Hep3B and HepG2 were detected by qRT-PCR and Western blot.Human hepatoma cells HepG2 cells were divided into control group,NC-mimics group,miR-508-3p mimics group,miR-508-3p mimics+si-NC group and miR-508-3p mimics+si-CLEC1B group.Colony for-mation assay and Transwell assay were used to detect cell proliferation and invasion abilities,respectively.Western blot was used to detect the expressions of Hedgehog signaling pathway-related proteins(Shh,Gli1 and Ptch1).Targetscan bioinformatics website was used to predict the downstream target genes of miR-508-3p,and the target relationship between miR-508-3p and CLEC1B was de-tected by dual luciferin assay.Results Compared with adjacent normal tissues,the expressions of miR-508-3p gene and CLEC1B gene were significantly decreased in liver cancer tissues(P<0.05),and the expressions of miR-508-3p and CLEC1B were positively correlated in liver cancer tissues(R2=0.746 2,P<0.001).Compared with control group and NC-mimics group,the proliferation and in-vasion abilities of HepG2 cells were significantly decreased in miR-508-3p mimics group(P<0.05),and the protein expressions of Shh,Gli1 and Ptch1 in Hedgehog signaling pathway were decreased(P<0.05).Dual luciferase reporter gene assay confirmed that CLEC1B was the target of miR-508-3p.The response test showed that compared with miR-508-3p mimics+si-NC group,the cell proli-feration and invasion abilities were significantly increased in miR-508-3p mimics+si-CLEC1B group(P<0.05),and the protein expres-sions of Shh,Gli1 and Ptch1 were increased(P<0.05).Conclusion Overexpression of miR-508-3p can target to increase CLEC1B expression and inhibit Hedgehog pathway proteins,thereby inhibiting proliferation and invasion of HepG2 cells.
