SALL4对宫颈癌细胞顺铂耐药性的影响及其机制
Effect and mechanism of SALL4 on cisplatin resistance in cervical cancer

Abstract
Objective To explore the role and mechanism of spalt-like transcription factor 4(SALL4)in cisplatin(DDP)resistance of cervical cancer.Methods The overexpression plasmid of SALL4(pCAG-IRES2-AcGFP1-Neo-SALL4)was constructed,then the overexpression plasmid and empty plasmid(pCAG-IRES2-AcGFP1-Neo)were transfected respectively into HeLa and SiHa cells,and the cells stably expressing SALL4 were screened by neomycin(G418).The cisplatin resistant cell lines HeLa-DDP and SiHa-DDP were established using concentration gradient increasing method.The cervical cancer cells in SALL4 overexpression group and control group were cultured with different concentrations of cisplatin(3,6,12,24,48 μg/mL)for 48 h,then the half maximal inhibitory con-centration(IC50)of cisplatin was calculated using MTT assay.The cells in SALL4 overexpression group and control group were cul-tured with cisplatin(5 μg/mL)for 24,48,72 h,and the cell viability was calculated using MTT assay.The mRNA and protein expres-sion levels of SALL4,ABCG2,and MDR1 in SALL4 overexpression cells and cisplatin resistant cell lines were detected by qRT-PCR and Western blot,respectively.Results After 48 h treatment,the cell viability in SALL4 overexpression group was higher than that in control group under different concentrations of cisplatin,and the IC50 of cisplatin was higher(P<0.05).Under the same concentra-tion of cisplatin,the cell viability in SALL4 overexpression group was significantly higher than that in control group at 48 h and 72 h(P<0.05).Compared with control group,the mRNA and protein expression levels of ABCG2 and MDR1 were significantly increased in SALL4 overexpression group(P<0.05).The mRNA and protein expression levels of SALL4,ABCG2,and MDR1 in HeLa-DDP and SiHa-DDP cells were significantly higher than those in parental cells(P<0.05).Conclusion Overexpression of SALL4 can induce the resistance of human cervical cancer cells to cisplatin,which may be related to the regulation of cell resistance-related genes ABCG2 and MDR1.关键词
