全蝎软膏对皮肤烫伤创面愈合的疗效及其机制
Efficacy and its mechanism of Scorpion Ointment on the healing of scald wounds

Abstract
Objective To investigate the potential therapeutic effects and underlying mechanism of Scorpion Ointment on the healing of deep second-degree scald wounds in a rat model.Methods SPF-grade 6-8-week-old male SD rats were randomly divided into four groups:blank control group,model group,Moist Burn Ointment group,and Scorpion Ointment group,with 10 rats in each group.A deep second-degree scald rat model was established using a heated weight.After successful modeling,the rats subjected to a 2 mL nor-mal saline-soaked dressing in model group,and the rats were treated with topical application of Scorpion Ointment and Moist Burn Ointment in Scorpion Ointment group and Moist Burn Ointment group,respectively.The ointments were applied evenly to a thickness of about 2 mm on the scald wound surface,twice daily for 15 consecutive days.The rats in blank control group underwent only depila-tion without modeling.The healing of the scald wounds was observed in each group,and the wound healing rate was calculated.Histo-pathological changes in the scalded skin tissues were examined using HE staining and Masson staining.The levels of TNF-α,IL-1β,and vascular endothelial growth factor(VEGF)in the scalded skin tissues were measured using ELISA.Results Compared with blank control group,the wound healing rate was significantly decreased in model group(P<0.05),the skin tissue was markedly thinned and necrotic,accompanied by numerous inflammatory cell infiltrations,only a small amount of connective tissue and neovas-cularization was observed,and the collagen fibers were disorganized.Compared with blank control group,the levels of TNF-α and IL-1β in the scalded skin tissues were significantly increased in model group(P<0.05),while the level of VEGF was significantly de-creased(P<0.05).Compared with model group,the scald wound area was significantly reduced in Moist Burn Ointment group and Scorpion Ointment group,and the wound healing rate was significantly increased(P<0.05);the epidermal cells of the skin tissue showed significant hyperplasia,forming a thicker layer of new epidermis,a large number of fibroblasts were observed in the dermis,and both connective tissue and neovascularization were significantly increased with well-organized collagen fibers;the levels of TNF-α and IL-1β in scalded skin tissue were significantly decreased(P<0.05),while the level of VEGF was significantly increased(P<0.05).Conclusion Scorpion Ointment can promote the healing of deep second-degree scald wounds in rats by inhibiting inflammatory re-sponses and promoting neovascularization.关键词
