降钙素受体样受体对子痫前期胎盘滋养细胞增殖和侵袭的影响
Effect of calcitonin receptor-like receptor on proliferation and invasion of placental trophoblastic cells in preeclampsia

Abstract
Objective To explore the effect of calcitonin receptor-like receptor(CRLR)on proliferation and invasion of placental tro-phoblastic cells in preeclampsia.Methods Human chorionic trophoblast cells(HTR-8/SVneo cells)were divided into NC group,H/R group,H/R+NC-oe group,H/R+CRLR-oe group,H/R+NC-sh group and H/R+CRLR-sh group.HTR-8/SVneo cells in NC group were routinely cultured for 48 h.After verifying transfection efficiency by qRT-PCR and Western blot,HTR-8/SVneo cells in H/R+NC-oe group,H/R+CRLR-oe group,H/R+NC-sh group and H/R+CRLR-sh group were treated with hypoxia/reoxygenation(H/R)for 48 h.The relative cell viability was detected by CCK-8 method,the EdU positive rate of cells was detected by EdU staining,the apoptosis rate(TUNEL positive rate)was detected by TUNEL staining,and the number of cell invasion was detected by Transwell method.Intra-cellular reactive oxygen species(ROS)concentration was measured using DCFH-DA,malondialdehyde(MDA)content was detected by the thiobarbituric acid(TBA)method,superoxide dismutase(SOD)activity was assayed by WST-8 method,and glutathione peroxi-dase(GSH-Px)activity was determined by DTNB method.CRLR mRNA level was analyzed by qRT-PCR,and the protein expressions of CRLR,apoptosis-related proteins(Bcl-2,cleaved Caspase-3),anti-apoptotic pathway proteins(p-PI3K,PI3K,p-Akt,Akt),and invasion-related proteins(MMP2 and MMP9)were detected by Western blot.Results Compared with NC group,the relative cell vi-ability,EdU positive rate and the number of invasion cells were decreased in H/R group(P＜0.05),the TUNEL positive rate was in-creased(P＜0.05),the protein expressions of Bcl-2,MMP2 and MMP9 were decreased while cleaved Caspase-3 protein level was in-creased(all P＜0.05),the relative fluorescence intensity of ROS and MDA level were increased(P＜0.05),SOD and GSH-Px levels were decreased(P＜0.05),and the relative phosphorylation levels of PI3K and Akt proteins were decreased(P＜0.05).Compared with H/R+NC-oe group,the relative cell viability,EdU positive rate and the number of invasion cells were increased in H/R+CRLR-oe group(P＜0.05),the TUNEL positive rate was decreased(P＜0.05),the protein levels of Bcl-2,MMP2 and MMP9 were increased while cleaved Caspase-3 protein level was decreased(all P＜0.05),the relative fluorescence intensity of ROS and MDA level were de-creased(P＜0.05),SOD and GSH-Px levels were increased(P＜0.05),and the relative phosphorylation levels of PI3K and Akt proteins were increased(P＜0.05).Compared with H/R+NC-sh group,the relative cell viability,EdU positive rate and the number of invasion cells were decreased in H/R+CRLR-sh group(P＜0.05),the TUNEL positive rate was increased(P＜0.05),the protein levels of Bcl-2,MMP2 and MMP9 were decreased while cleaved Caspase-3 protein level was increased(all P＜0.05),the relative fluorescence inten-sity of ROS and MDA level were increased(P＜0.05),SOD and GSH-Px levels were decreased(P＜0.05),and the relative levels of PI3K and Akt protein phosphorylation were decreased(P＜0.05).Conclusion Upregulation of CRLR can promote the proliferation and invasion of preeclampsia cells,and inhibit the cell apoptosis,which is related to the activation of the PI3K/Akt signaling pathway.关键词
