摘要

Abstract

In this study,we screened the sequences with high nucleic acid variability as candidate DNA barcodes through the plastid genome of Glycyrrhiza,amplified and sequenced them by PCR,and combined with the sequence data from the GenBank database to evaluate the identification ability of different DNA barcodes by phylogenetic tree construction and genetic distance analysis.The results showed that among the 10 candidate DNA barcodes screened,trnF-trnL,trnQ-accD and trnR-trnN had the highest identification success rate,which could separate seven species of the genus Glycyrrhiza,and the minimum interspecific variation was greater than the maximum intraspecific variation in all three sequences and the minimum intraspecific variation was observed.The combination of trnF-trnL,trnQ-accD and trnR-trnN sequences resulted in a 100%identification success rate,which can be used as the optimal sequence combination for accurate identification of Glycyrrhiza.The present study provide a scientific basis for the molecular identification of the genus of Glycyrrhiza.
