Abstract
Objective To investigate the effect of long non-coding RNA H19 on the epithelial mesenchymal transition(EMT)process of oral squamous cell carcinoma Tca8113 cells through targeted regulation of microRNA-107(miR-107).Methods Obtain tumor tissue and normal tissue adjacent to oral squamous cell carcinoma patients,and use real-time fluorescence quantitative polymerase chain reaction(RT qPCR)to detect the expression levels of lncRNA H19 and miR-107.Tca8113 cells were cultured in vitro and divided into control group(conventional cell culture without any intervention treatment),H19 interference negative control group(transfected with siRNA without specific targeting relationship with H19),H19 interference group(transfected with siRNA specifically targeting H19),miR-107 overexpression negative control group(transfected with a mimic without specific targeting relationship with miR-107),miR-107 simulation group(transfected with miR-107 mimic),miR-107 inhibition negative control group(transfected with a mimic without specific targeting relationship with miR-107),and miR-107 inhibition group(transfected with miR-107 inhibitor).Use RT qPCR to detect the levels of lncRNAH19 and miR-107,Transwell assay to detect the invasion ability of Tca8113 cells,cell scratch assay to detect the migration ability of Tca8113 cells,RT qPCR to detect the levels of E-Cadherin,N-Cadherin,Vimentin and Snail.Results The relative expression levels of lncRNA H19 in tumor tissue and adjacent tissue of patients with oral squamous cell carcinoma were 1.56±0.42 and 1.06±0.28,respectively,the relative expression levels of miR-107 were 0.42±0.11 and 1.03±0.27,respectively,with statistically significant differences(P<0.05).The number of invasive cells of control group,H19 interference negative control group,H19 interference group,miR-107 overexpression negative control group,miR-107 simulation group,miR-107 inhibition negative control group and miR-107 inhibition group was(462.60±43.66)、(459.90±39.68)、(393.50±33.32)、(463.60±44.78)、(589.40±61.05)、(460.30±44.14)and(363.50±33.77),respectively.The cell migration rates were(33.54±3.06)%,(32.67±5.86)%,(25.18±2.58)%,(34.93±4.28)%,(19.52±2.28)%,(36.75±4.87)%and(53.61±6.03)%,respectively.The above indicators of the H19 interference group and miR-107 simulation group were compared with the control group,and the above indicators of the miR-107 inhibition group were compared with the control group,and the differences were statistically significant(all P<0.05).The relative expression levels of lncRNA H19 were 1.04±0.13,1.02±0.09,0.37±0.04,1.07±0.09,1.02±0.10,0.99±0.11 and 1.08±0.11,respectively.The relative expression levels of miR-107 were 1.03±0.10,1.06±0.11,1.87±0.12,1.07±0.16,1.56±0.18,1.03±0.14 and 0.42±0.08,respectively.The relative expression levels of E-Cadherin mRNA were 0.85±0.18,0.86±0.21,1.67±0.35,0.84±0.32,1.78±0.40,0.83±0.42 and 0.61±0.18,respectively.The relative expression levels of N-Cadherin mRNA were 1.65±0.46,1.69±0.42,1.11±0.38,1.62±0.46,1.07±0.33,1.66±0.53 and 2.08±0.53,respectively.The relative expression levels of Vimentin mRNA were 1.78±0.52,1.73±0.46,1.09±0.32,1.65±0.51,0.96±0.31,1.72±0.57 and 2.08±0.65,respectively.The relative expression levels of Snail mRNA were 1.37±0.33,1.38±0.38,0.98±0.27,1.36±0.32,0.77±0.19,1.39±0.36 and 1.86±0.46,respectively.The above indicators of the H19 interference group and miR-107 simulation group were compared with the control group,and the above indicators of the miR-107 inhibition group were compared with the control group,and the differences were statistically significant(all P<0.05).Conclusion lncRNA H19 promotes EMT and enhances the invasion and migration ability of oral squamous cell carcinoma Tca8113 cells by downregulating miR-107 expression.关键词
