佛手柑内酯对炎症刺激的牙髓干细胞成骨分化的研究
Research of bergapten on the osteogenic differentiation of dental pulp stem cells stimulated by inflammation

Abstract
Objective To explore the mechanism of bergapten(BeG)regulating CXC chemokine ligand-14(CXCL14)mediated macrophage polarization on osteogenic differentiation of dental marrow stem cells stimulated by inflammation based on phosphatidylinositol 3 kinase/protein kinase B(PI3 K/AKT)signaling pathway.Methods Human dental pulp stem cells(HDPSC)were randomly divided into control group(general culture),model group[10 μg·mL-1 lipopolysaccharides(LPS)],experimental group(15 μmol·L-1BeG+10 μg·mL-1 LPS)and si-CXCL14 group(transfected with si-CXCL14 based on experimental group).Indirect co-culture of HDPSC with tohoku hospital pediatrics-1(THP-1)were divided into THP-1+control group(untreated HDPSC co-cultured with THP-1),THP-1+model group(untreated HDPSC co-cultured with lower chamber THP-1+LPS),THP-1+experimental group(HDPSC treated with 15 μmol·L-1 BeG was co-cultured with THP-1+LPS)and THP-1+si-CXCL14 group(transfected with si-CXCL14 based on THP-1+experimental group).Reverse transcription real-time fluorescence quantitative polymerase chain reaction detected the messenger RNA(mRNA)expression level of CXCL14,and alkaline phosphatase(ALP)staining detected the changes in ALP activity of the cells in all groups after osteogenic induction.The protein relative expression levels of macrophage polarization markers and osteoblastic differentiation markers were detected by Western blot after administration.Results The relative mRNA expression levels of CXCL14 in control group,model group and experimental group were 1.00±0.12,0.63±0.08 and 0.87±0.14,respectively;the ALP scores after osteogenic induction were(3.25±0.66),(0.75±0.43)and(2.88±0.60)points,respectively;the ALP scores in si-CXCL14 group was(1.88±0.60)points,there were statistically significant differences in the relative mRNA expression levels of CXCL14 and ALP scores between model group and control group,and between experimental group and model group,there were statistically significant differences in ALP scores between si-CXCL14 group and experimental group(all P<0.01).In THP-1+control group,THP-1+model group,THP-1+experimental group and THP-1+si-CXCL14 group,inducible nitric oxide synthase(iNOS)relative expression levels were 1.00±0.15,1.92±0.27,1.23±0.19 and 1.74±0.26,respectively;the relative expression levels of arginase1(Arg-1)were 1.00±0.16,0.45±0.06,0.85±0.13 and 0.66±0.10,respectively;the relative expression levels of osteocal-cin(OCN)were 1.00±0.13,0.74±0.09,0.92±0.14 and 0.77±0.11,respectively;THP-1+model group compared with THP-1+control group,THP-1+experimental group compared with THP-1+model group,and THP-1+si-CXCL14 group compared with THP-1+experimental group were all significantlly differented in iNOS,Arg-1 and OCN relative expression levels(all P<0.05).Conclusions BeG affects macrophage polarization by regulating CXCL14,thus promoting osteogenic differentiation of HDPSC under inflammatory stimulation,which may be related to the regulation of PI3K/AKT pathway.关键词
