Effects and Mechanisms of Runmu Ling Formula on Corneal Epithelial Cell Pyropto-sis in a Rat Model of Dry Eye

摘要

Abstract

OBJECTIVE To investigate the effects and underlying mechanisms of Runmu Ling Formula on corneal epithelial cell pyroptosis in rat model of dry eye.METHODS A total of 40 rats were subcutaneously injected with 2 mg/g scopolamine hydrobromide solution to establish dry eye model and were randomly assigned to the model group(MG),low-dose Runmu Ling Formula group(L-RML),high-dose Runmu Ling Formula group(H-RML),and NOD-like receptor protein 3(NLRP3)inflammasome inhibitor group(MCC950),with an additional control group(CG),ten rats in each group.The L-RML and H-RML groups received daily intragastric administration of Runmu Ling Formula at 5.25 g/kg and 10.50 g/kg,respectively;the MCC950 group received intraperitoneal injection of MCC950 solution at 10 mg/kg once daily,for 14 consecutive days.Before and after modeling,Schirmer's Ⅰ test(SⅠT),tear film break-up time(BUT),and corneal fluorescein staining(FL)were performed.After treatment,corneal ultrastructure was observed by transmission electron microscopy,and the mRNA and protein expression of NLRP3,apoptosis-associated speck-like protein(ASC),Caspase-1,gasdermin D(GSDMD),and interleukins(IL)were detected.RESULTS(1)SⅠT:On days 10 and 24,MG had significantly lower SⅠT than CG.On day 24,L-RML,H-RML,and MCC950 groups had higher SⅠT than MG(all P<0.05).(2)BUT:On days 10 and 24,MG had shorter BUT than CG.On day 24,H-RML and MCC950 groups showed prolonged BUT compared with MG(all P<0.05).(3)FL score:On days 10 and 24,MG showed higher FL scores than CG.On day 24,H-RML and MCC950 groups exhibited significantly lower scores than MG(all P<0.05).(4)Pyroptosis-related mRNA:MG showed higher corneal NLRP3,GSDMD,ASC,and Caspase-1 mRNA expression than CG.L-RML,H-RML,and MCC950 groups had lower expression than MG,with MCC950 showing lower NLRP3 mRNA expression than L-RML(all P<0.05).(5)Pyroptosis-related proteins:MG exhibited higher NLRP3,GSDMD,ASC,and Caspase-1 protein expression than CG.H-RML and MCC950 groups showed reduced expression compared with MG,with MCC950 presenting lower NLRP3,GSDMD,and ASC expression than L-RML,and lower GSDMD expression than H-RML(all P<0.05).(6)Inflammation-related proteins:MG demonstrated increased IL-1β and IL-18 expression compared with CG.H-RML and MCC950 groups showed reduced expression compared with MG,and MCC950 was lower than L-RML(all P<0.05).CONCLUSIONS Runmu Ling Formula may improve tear film homeostasis in rats with dry eye by inhibiting NLRP3 inflammasome activation,downregulating Caspase-1/GSDMD-mediated corneal epithelial cell pyroptosis,and reducing the release of inflammatory cytokines such as IL-1β and IL-18.
