摘要

Abstract

The aim of this study was to investigate the effect of geometric orifice cavitation on drug encapsulation in chitosan nanoparticles.5-Fluorouracil(5-Fu)was used as a model drug.The effects of pore structure,inlet pressure,oil/water ratio,HLB value and crosslinking time of compound emulsifier on encapsulation efficiency of chitosan nanoparticles were investigated.5-Fu chitosan nanoparticles were characterized.The results show that the pore size of 3.0 mm is more favorable for the preparation of nanoparticles with good encapsulation performance,and the encapsulation efficiency of nanoparticles prepared by porous plates with a smaller open porosity and longer circumference is higher than that by porous plates.Moreover,for orifice plates with the same open area ratio,those with a larger specific perimeter also result in nanoparticles with higher encapsulation rates.As the inlet pressure,oil/water ratio,HLB value and crosslinking time of compound emulsifier increase,the encapsulation efficiency initially increases and then decreases.FTIR analysis shows that after cavitation,5-Fu is successfully loaded into chitosan nanoparticles without any changes in its functional groups,indicating no drug interaction between 5-Fu and chitosan nanoparticles.
