摘要

Abstract

In order to explore the biological characteristics,genomes feature of Salmonella phages,the ex-periment isolated phages from dairy farms in Lanzhou city.The double-layer agar plate method was used for the purification,ultracentrifugation method was used for the concentration,and Transmission electron microscopy was used to observe the phage morphology.Biological characteristics such as phage host spec-trum,one-step growth curve,optimal infection multiplicity,acid-base and thermal stability were deter-mined,and phage genomes were extracted for sequencing and analysis.The results showed that one strain of Salmonella paratyphi C phage was isolated and named SP2,and the morphological observations show that the SP2 head is polyhedral with a diameter of 70 nm and a constricted tail of approximately 130 nm.The incubation period was 40 min,the outbreak period was 40~200 min,the optimal infection multiplicity was 0.001,and the activity was good at the temperature≤50 ℃ and the pH value was 4~11.The total length of the sp2 genome was 85 702 bp,containing 124 ORFs,only 36 ORFs were known coding genes,containing 21 tRNAs,and did not carry drug resistance genes,virulence genes and lysogenic genes.The bio-logical characteristics of SP2 are stable and the genetic background is clear,in order to lay a realistic foun-dation for future phage therapy.
