Abstract

[Objective]The effects of seed-soaking temperature,cultivation temperature and salt concentration on seed germination and seedling physiological indexes of Glycyrrhiza uralensis were explored to provide a theoretical basis for seedling cultivation and field management.[Method]The germination experiments of G.uralensis seeds were conducted to compare the effects of different temperatures,and NaCl and Na2SO4 at varying concentrations on seed germination,seedling growth indexes,and physiological indexes.[Result]Seed germination was significantly influenced by temperature,with optimal results observed at 25 ℃for both seed soaking and cultivation.Both NaCl and Na2SO4 followed similar trends,showing low concentrations promoted germination,while high concentrations inhibited it.45 mmol/L NaCl and Na2SO4 significantly enhanced seed germination,with NaCl showing a stronger promotive effect.These treatments also significantly increased radicle growth(but not hypocotyl width)and elevated MDA and Pro content,alongside boosting CAT,POD,and SOD activities,thereby improving seedling growth.Under NaCl stress,radicle length and hypocotyl width were negatively correlated with MDA content,Pro content,and POD activity,but positively correlated with SOD activity.Under Na2SO4 stress,both radicle length and hypocotyl width showed significant negative correlations with Pro content and POD activity,and negative correlations with MDA content,CAT activity and SOD activity.[Conclusion]G.uralensis seeds should be soaked and cultured at 25℃.Under this condition,45 mmol/L NaCl and Na2SO4 stress had the best performance on G.uralensis seed germination index,seedling radicle growth and physiological index,which can be popularized and applied in G.uralensis seed seedling.
