Abstract

[Objective]The effects of 60Co-γ radiation on germination,emergence and physiological indexes of Perilla frutescens seeds were investigated to determine the suitable radiation dose range and provide a theoretical basis for radiation mutation breeding of P.frutescens.[Method]The germination rate,germination index,germination potential,seed vigor index,seedling emergence rate,seedling height,main root length,fresh seedling weight,seedling vital index,SOD,POD and CAT activity and MDA content of Guisu 3 seeds(a P.frutescens variety)radiated by 60Co-γ with 100-500 Gy were determined to analyze the effects of different radiation doses on P.frutescens seeds by taking untreated seeds as the control.[Result]The germination rate,germination index,germination potential and vigor index of P.frutescens seeds radiated by 60Co-γ showed a decreasing trend with increase of radiation dose and the germination rate,germination index and germination potential were no significant differences compared with CK when the seeds radiated by 60Co-γ with 100-200 Gy.The germination rate,germination index,germination potential and vigor index of P.frutescens seeds radiated by 60Co-γ with 300-500 Gy showed significant differences compared with CK and the germination rate,germination index,germination potential and vigor index of P.frutescens seeds radiated by 60Co-γ with 500 Gy were all the lowest with 4.22%,0.81,2.44%and 0.12 respectively.The seedling emergence rate,seedling height,mainroot length,fresh seedling weight and seedling vigor index all showed a deceasing trend with increase of radiation dose and the seedling emergence was completely inhibited when the radiation dose was 500 Gy.The seedling emergence rate,seedling height,mainroot length,fresh seedling weight and seedling vigor index of P.frutescens seeds radiated by 60Co-γ with 100-400 Gy were 6.75%-95.00%,18.22%-75.70%,6.59%-76.50%,7.13%-69.95%and 11.68%-99.07%lower than CK respectively.The seedling emergence rate,mainroot length,fresh seedling weight and seedling vigor index of P.frutescens seeds radiated by 60Co-γ with 100 Gy had no significant differences compared with CK.The seedling emergence rate,seedling height,mainroot length,fresh seedling weight and seedling vigor index of P.frutescens seeds radiated by 60Co-γ with 200-400 Gy showed significant differences compared with CK.The LD50 and LD40 of P.frutescens seeds were 218.92 Gy and 271.55 Gy respectively.The seedling SOD,POD and CAT activity showed a rising first and then decreasing trend with increase of radiation dose,and the SOD,POD and CAT activity of seedlings from P.frutescens seeds radiated by 60Co-γ with 300 Gy reached 63.11 U/g,16.12 U/g and 8.11 U/g respectively.The MDA content in seedlings showed a rising trend with increase of radiation dose and the MDA content in seedlings from P.frutescens seeds radiated by 60Co-γ with 100-400 Gy was 15.38%-102.56%higher than CK significantly.The correlation analysis showed that the radiation dose was negatively related to emergence rate,seedling height,seedling vigor index,mainroot length significantly or very significantly,and positively related to SOD activity and MDA content of seedlings significantly.[Conclusion]The suitable radiation dose range for Guisu 3 is 218.92-271.55 Gy.The damage degree of P.frutescens seeds caused by 60Co-γ radiation is related to MDA content.
