Abstract

[Objective]The correlation between polymorphisms of the transmembrane protein 68 gene(TMEM-68)and growth traits in Wuchuan black cattle were studied,which provided references for screening the domain group of Wuchuan black cattle and protecting the germplasm resource of cattle.[Method]Taking Wuchuan black cattle as study object,PCR and lluminaX-10 high-throughput sequencing were employed to obtain the complete sequence of the TMEM-68 gene in Wuchuan black cattle.Futhermore,the single nucleotide polymorphisms(SNPs)in the TMEM-68 gene associated with growth traits were analyzed,and the superior allelic variants were explored.[Result]8 SNP sites significantly associated with growth traits were detected,including 224(T/A),360(G/A),447(T/C),557(G/T),938(A/G),1135(A/G),1319(G/A),and 1567(T/A)loci of TMEM-68 gene in Wuchuan black cattle.The polymorphic loci of 8 SNP sites owned a high degree of homozygosity(0.65-0.71),showing low polymorphism(PIC<0.25).At 224(T/A)site,the chest width and body weight of TT genotype individual were significantly higher than those of TA and AA.At the 360(G/A)site,the height of cross section,chest circumference and body height of AA genotype individual was significantly than GA.At the 447(T/C)site,the body height,height of cross section and chest width of CC genotype individual were significantly higher than TT.At the 557(G/T)site,the body diagonal length of GT and TT genotype individual were significantly higher than GG.At the 938(A/G)site,the chest width of GG and AA genotype individual were obviously higher than AG.At the 1135(A/G)site,the pin bone width of AA and GG genotype individual were significantly higher than AG.At the 1319(G/A)site,the height of cross section of GA and AA genotype individual were obviously higher than GG.At the 1567(T/A)site,the chest width of TT and TA were significantly than AA.Haplotype analysis demonstrated that there were 7 haplotypes in Wuchuan black cattle population,among which the number of haplotypes of H1 were significantly more than other haplotype combinations,indicating that it was the dominant haplotype.The analysis about haplotypes combination showed that H2H6 haplotype had great advantages in the height of cross section,body diagonal length,canon circumference and width of pin bone,and H1H7 haplotype had advantages in chest width,body height and body weight.[Conclusion]TMEM-68 gene was closely related to the growth traits of Wuchuan black cattle,could serve as a candidate molecular marker for the development and breeding of genetic resources of Wuchuan black cattle.
