Expression of CHST7 and SHH in invasive non-functioning pituitary neuroendocrine tumors and their association with clinicopathological features

Abstract
Objective:Identifying biomarkers to predict the invasiveness of non-functioning pituitary neuroendocrine tumors(PitNET)to guide early clinical management is a current research focus.Carbohydrate sulfotransferase(CHST)7 and Sonic hedgehog(SHH)are associated with the occurrence and progression of various tumors.This study aims to investigate the expression of CHST7 and SHH in invasive non-functioning PitNET and their relationship with clinicopathological features. Methods:A retrospective analysis was conducted on 52 patients with non-functioning PitNET treated at the Neurosurgery Department of Tangshan People's Hospital.Based on preoperative MRI and Knosp classification,patients were divided into an invasive group(n=28)and a non-invasive group(n=24).Expression levels of CHST7 and SHH in serum and tumor tissues were detected by enzyme-linked immunosorbent assay(ELISA)and immunohistochemistry.Their associations with clinicopathological features(Ki-67,p53,and lineage-specific transcription factor expression)were further analyzed. Results:Preoperatively,serum CHST7 and SHH protein levels in the invasive group were significantly higher than those in the non-invasive group(both P<0.05).Three weeks postoperatively,serum CHST7 and SHH levels decreased in both groups but remained significantly higher in the invasive group(both P<0.05).In invasive non-functioning PitNET,patients with Ki-67≥3%had higher CHST7 and SHH positivity than those with Ki-67<3%(both P<0.05).Strong p53-positive patients also showed higher CHST7 and SHH positivity compared to weak or negative p53 patients(both P<0.05).CHST7 positivity in patients with transcription factor 19(T-PIT)expression was significantly higher than that in patients without,while SHH positivity in patients with pituitary-specific POU-class homeodomain transcription factor-1(PIT1)expression was significantly higher than that in patients without(both P<0.05). Conclusion:High expression of CHST7 and SHH may be associated with the invasiveness of non-functioning PitNET.关键词
