Improvement of agronomic and quality traits of Yangmai 18 using CRISPR/Cas9 gene editing technology

Abstract
Jiangsu province is an important wheat-producing province in China,spanning the middle and lower reaches of the Yangtze River(MLRYR)and the Huang-Huai region,two major wheat-growing areas.The wheat planted in the MLRYR is mainly red-skinned spring wheat,which usually has large panicles and a loose plant type,and is resistant to Fusarium head blight(FHB),powdery mildew(PM),and pre-harvest sprouting(PHS).In contrast,the wheat planted in the Huang-Huai re-gion is mainly white-skinned winter wheat,which has a compact plant type and a higher number of panicles,but is generally sus-ceptible to FHB,PM,and PHS.To develop new wheat germplasms that meet different application needs,the vernalization-relat-ed gene Vrn-D1,grain hardness-related genes Pina and Pinb,and the seed coat color-related gene Myb10 were edited in the red-skinned spring soft wheat variety Yangmai 18 using CRISPR/Cas9 technology.A total of six Vrn-D1 mutants,five Pin mutants,and seven Myb10 mutants were obtained.Phenotypic identification results showed that compared with the wild type,the second and fourth internode lengths of the Vrn-D1 mutants were significantly reduced(with the exception of the vrnM5 line),the grain hardness of the Pin mutants was sig-nificantly increased,and the seed coat color of five Myb10 mutants changed from red to white.The results of this study provide a reference for using CRISPR/Cas9 gene editing technology to improve specific agronomic and quality traits of wheat.关键词
