参芪颗粒通过调控p70S6K蛋白降低自噬水平治疗C-BSA诱导的膜性肾病大鼠的作用机制
Mechanism of Shenqi Moshen Granules(参芪膜肾颗粒)in Treatment of C-BSA-induced Membranous Nephropathy in Rats by Decreasing Autophagy Level through Regulating p70S6K Protein

Abstract
Objective To investigate the mechanism of Shenqi Moshen Granules(参芪膜肾颗粒,SQ)on membranous ne-phropathy by regulating the level of renal autophagy.Methods Cationic bovine serum albumin(C-BSA)was used to induce a rat model of membranous nephropathy(SD),and the experimental groups were divided into normal(N),model(M),cyclosporine(CsA),and traditional Chinese medicine(SQ)groups after successful modeling.after 4 weeks of intervention,urinary protein was detected in each group.PAS staining was performed to view the pathology of the kidney,and the distribution of F-actin was de-tected in the kidneys with the application of immunofluorescent technique,and protein blotting was performed to detect F-actin in the kidneys.The distribution of F-actin in the kidneys was examined by PAS staining,and the expressions of Synaptopodin,p70S6K,autophagy-related protein p62 and vps34 were detected by protein blotting.Results Compared with those in the group N,the rats in group M showed significant proteinuria,and PAS pathology revealed thickening of glomerular basement membrane,reduced expression of renal synaptic protein Synaptopodin,decreased fluorescent expression of F-actin,and increased expressions of autophagy-associated proteins p62 and vps34.Compared with the M group,the SQ group and CsA group had lower proteinuri-a,reduced glomerular basement membrane damage,elevated Synaptopodin expression,elevated F-actin fluorescence expression,and elevated expressions of p70S6K,p62 and vps34.Conclusion SQ can reduce proteinuria and attenuate renal injury in C-BSA rats.Its mechanism of action may be realized by increasing the expression of p70S6K protein,decreasing the autophagy proteins of p62 and vps34,and maintaining the stabilization of synaptic protein Synaptopodin and skeletal microfilament F-actin.关键词
