针刺天枢穴联合中频电刺激改善脊髓损伤后肠道功能障碍的作用机制
Mechanism of Acupuncture at "Tianshu(ST25)" Combined with Medium-Frequency Electrical Stimulation in Improving Intestinal Dysfunction after Spinal Cord Injury

Abstract
Objective To observe the improvement of autonomic dysfunction in rats with T10 spinal cord contusion injury trea-ted with acupuncture at "Tianshu(ST25)" combined with medium-frequency electrical stimulation(MFEs)and to explore its mechanism of affecting intestinal function.Methods In this study,40 female SD rats were divided into four groups:a blank group,a model group,an MFES group,and an MFES combined with acupuncture(MFES+Acu)group,with 10 rats in each group.The T10 spinal cord contusion injury model was established using the modified Allen's WD method.The MFES group received bilater-al MFES at "Tianshu(ST25)",while the MFES+Acu group received additional acupuncture.Evaluations included hindlimb mo-tor function,intestinal function recovery time,and biomarker expression in spinal cord and colonic tissues among groups,to com-prehensively investigate the therapeutic effects of "Tianshu(ST25)" combined with MFES treatment.Results Compared with those in the model group,the rats in the MFES group and MFES+Acu group showed significantly improved motor function(P<0.05),with the latter demonstrating superior outcomes(P<0.05).In terms of intestinal function,the first melena excretion time was reduced in both the MFES group and MFES+Acu group(P<0.0001).Compared to the model group,the MFES+Acu group exhibited further optimization(P<0.001).In spinal cord tissue,neurokinin-1 receptor(NK1R)expression decreased in the MFES group(P<0.05)and both substance P and NK1R decreased in the MFES+Acu group compared to those in the model group(P<0.05).Compared to the MFES group,the MFES+Acu group had reduced LC3B and increased p62(P<0.01).In co-lonic tissue,substance P was reduced in the MFES+Acu group compared to that in both the model group(P<0.0001)and MFES group(P<0.001).Additionally,NK1R expression decreased in both the MFES group(P<0.05)and MFES+Acu group(P<0.0001)compared to that in the model group,with a more pronounced decrease in the latter(P<0.001).Conclusion Acu-puncture at "Tianshu(ST25)" combined with MFES can significantly improve neurogenic intestinal dysfunction after spinal cord injury by regulating the substance P/NK1R pathway and autophagy processes.Moreover,this combined treatment was more effec-tive than MFES alone at "Tianshu(ST25)".关键词
