摘要

Abstract

Objective To explore the potential mechanism of Yishen Tongluo Formula(益肾通络方,YSTLF)in treating oli-goasthenozoospermia with kidney deficiency and collateral obstruction.Methods Seventy-two SPF SD male rats were randomly divided into normal control(NC)group,glucosides of Tripterygium wilfordii Hook.F.(GTW)+adrenaline hydrochloride(A)group,GTW+A+YSTLF-low dose(L)group,GTW+A+YSTLF-middel dose(M)group,GTW+A+YSTLF-high dose(H)group and GTW+A+L-carnitine(LEV)group,12 rats in each group.All groups except NC group were given GTW sus-pension[40 mg·kg-1·d-1]by gavage at 10 o'clock from week 1 to week 4.In the 4th week of the experiment,the rats were given A injection[0.3 mg·kg-1·d-1]by subcutaneous injection after gavage of GTW,and then exposed to ice water for 10 min after 2 h of injecting every other day to establish the model of oligoasthenozoospermia with kidney deficiency and collateral ob-struction.From the 5 th to the 8th week of the experiment,at 10 o'clock every day,the rats in GTW+A+YSTLF-L group,GTW+A+YSTLF-M group and GTW+A+YSTLF-H group were given YSTLF suspension by gavage at 0.522,1.044,2.088 g·mL-1,respectively,and the rats in GTW+A+LEV group were treated with 18 mg·mL-1 levocarnitine suspension by gavage.After 8 weeks of experiment,the abdominal aorta blood and bilateral testis and epididymis tissues were picked out.The in-dexes included the general state,the sperm quality,coagulation function of the rats in each group.H&E staining was used to ob-serve the epididymis tissues pathological changes.Liquid chromatography-mass spectrometry was used to analyze testis metabolic profiles,screen differential metabolites among the groups and enrich related metabolic pathways.Results Compared with the GTW+A group,all the Yishen YSTLF groups could improve the TCM symptoms of kidney deficiency and collateral obstruction in the model rats.The GTW+A+YSTLF-H group and GTW+A+YSTLF-M group had significantly higher sperm motility and total sperm motility(P<0.05),and the GTW+A+YSTLF-H group also had a significantly higher sperm concentration(P<0.01).Prothrombin time(PT)was significantly prolonged(P<0.001)and fibrinogen(FIB)was decreased(P<0.05)in all YSTLF groups.thrombin time(TT)was prolonged in the GTW+A+YSTLF-M group and GTW+A+YSTLF-H group(P<0.05).Compared with the GTW+A group,YSTLF at all doses could significantly improve the epididymis tissues histopathologic injury and microvascular stasis was reduced.Metabolomics analysis results showed that there were 25 potential biomarkers in the testicular tissue of YSTLF in the treatment of oligoasthenozoospermia with kidney deficiency and collateral obstruction.It was mainly involved in the metabolism of methyl butyrate,D-glutamine and D-glutamate,glycerophospholipid metabolism,arginine and proline metabolism,arginine biosynthesis,cysteine and methionine metabolism,sphingolipid metabolism,aminoacyl-tRNA biosynthesis,and arachidonic acid metabolism etc.Conclusion YSTLF can improve the related symptoms and indicators of oligoas-thenozoospermia rat model with kidney deficiency and collateral obstruction.The mechanism of action may be related to regulating amino acid metabolism,lipid metabolism and energy metabolism.
