基于ASS1介导的Src/STAT3、MAPK/ERK级联反应探究参七虫草方对IPF模型大鼠成纤维细胞的影响
Probing Effects of Shenqi Chongcao Formula(参七虫草方)on Rat Fibroblasts in IPF Model Based on ASS1-mediated Src/STAT3 and MAPK/ERK Cascade Reaction

Abstract
Objective To investigate the effects of Shenqi Chongcao Formula(参七虫草方)on fibroblasts of bleomycin(BLM)-induced idiopathic pulmonary fibrosis(IPF)model rats.Methods Except for the blank group,the fibroblasts of the mod-el were isolated and cultured in vitro by intratracheal instillation of BLM.The proliferation of lung fibroblasts was detected by CCK-8 method,and the optimal concentration and time were selected for follow-up experiments.The experiment was divided into 6 groups:blank group[normal fibroblasts+normal drug-containing serum(group A)],model group[model fibroblasts+nor-mal drug-containing serum(group B)],best drug-containing serum group of Shenqi Chongcao Formula(group C),arginine succinate synthetase-1(ASS1)inhibitor group(group D),tyrosine kinase(Src)/signal transducer and transformation activator 3(STAT3)blocker group(group E),MAPK/extracellular regulatory protein kinase(ERK)pathway inhibitor group(group F).CCK-8 method and cell scratch assay were used to detect cell viability and migration rate,respectively.RT-qPCR was used to detect the expression levels of ASS1,Src,STAT3,bispecific protein kinase 1/2(MEK1/2)and ERK1/2.Western blot was used to detect the protein expression levels of p-ERK,activator-1(AP-1),P-MEK,matrix metalloproteinases(MMPs)and nuclear protein factor-κB p65 in each group.Results Compared with those in group A,cell viability and cell migration rate were en-hanced and increased in group B(P<0.05),while cell viability and cell migration rate in group B were decreased and decreased in group B(P<0.05).RT-qPCR results showed compared with those in group A,the expression levels of ASS1,MEK1,MEK2,ERK1,ERK2,Src and STAT3 genes were increased in group B(P<0.05).Compared with those in group B,the expression level of related genes in the other intervention groups decreased(P<0.05).The results of Western Blot showed that the protein expres-sion levels of p-ERK,P-MEK,MMP-2,MMP-9,NF-κB p65 and AP-1 in group B were increased compared with group A(P<0.05).Compared with those in group B,the expression levels of the other intervention groups decreased(P<0.05).Con-clusion Shenqi Chongcao Formula can inhibit ASS1-mediated Src/STAT3 and MAPK/ERK cascades,down-regulate the pro-tein expressions of MMP-2,MMP-9,AP-1 and NF-κBp65,and improve the progression of IPF.关键词
