七味白术散对2型糖尿病大鼠脂质代谢与炎性因子的作用研究
An Examination of the Effects of Seven Flavor Bai Zhu Powder on Lipid Metabolism and Inflammatory Biomarkers in Rats Diagnosed with Type 2 Diabetes

Abstract
Objective To elucidate the effects of Qiwei Baizhu San(QWS)on lipid metabolism and inflammatory factors in type 2 diabetes mellitus(T2DM)rats.Methods Six Sprague-Dawley(SD)rats were randomly selected as the control group,which was maintained on a normal diet.The remaining rats were subjected to a high-fat diet(HFD)for a duration of four weeks,after which they received A dosage of 30 mg/kg of streptozotocin(STZ)was administered via injection to create a T2DM model.Following the successful establishment of this model,the rats were randomly selected.selected from each group,which were cate-gorized into the model group(Model),the low-dose group of QWS(QWSL,8.775 g/kg),and the high-dose group of QWS(QWSH,17.55 g/kg),blood glucose and body weight were recordedadministered via gavage for a duration of four weeks.The his-topathological changes in rat liver tissue were observed via Hematoxylin and Eosin(HE)staining,and serum levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),free fatty acids(FFA),hormone-sensitive lipase(HSL),hepatic lipase(HL),lipoprotein lipase(LPL),adipose triglyceride lipase(ATGL),tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6)were detected by using ELISA.The mRNA expressions of PI3K,AKT and m-TOR in rat liver were detected by qRT-PCR and the protein expression levels of PI3K and AKT in rats were observed via immunohistochemical staining.The colorectal feces were collected from rats for 16SrDNA sequencing.Results Compared with the model group,the QWS groups showed lower blood glucose and higher body weight;significantly lower levels of liver function and related metabolic enzymes AST,ALT,FFA and HSL(P<0.05);significantly higher levels of HL,LPL and ATGL(P<0.05,P<0.01);and significantly lower levels of TNF-α and IL-6(P<0.05).The PCR results showed that the model group had a significantly reduce compared to the blank group(P<0.01),while the QWS group had a significant increase compared to the model group(P<0.05,P<0.01).The immu-nohistochemical results showed that the model group had a significant decrease compared to the blank group(P<0.01),while the high-dose QWS group had a significant increase(P<0.05).Furthermore,the analysis of intestinal flora indicated that the ad-ministration of QWS resulted in the improved intestinal dysbiosis.Compared to the model group,at the phylum level,there was an increase in the relative abundance of Thick-walled Bacillus and Anaplasma.Conversely,at the genus level,a decline in the a-bundance of Lactobacillus was observed.These flora changes were beneficial to the hypoglycaemic effect of QWS.Conclusion QWS can significantly ameliorate STZ-induced T2DM by exerting hypoglycemic effects through regulating inflammation caused by gut microbiota dysbiosis in rats.关键词
