摘要

Abstract

[Objective]This study aimed to explore the community structure and microbial diversity in winged and wingless Sitobion miscanthi.[Method]The study employed Illumina NovaSeq sequencing of the bacterial 16S rRNA gene(V3-V4 region)and fungal ITS1 region to investigate the differences in symbiotic microbial communities between winged and wingless fourth-instar nymphs and adults.[Result]A total of 737 705 high-quality bacterial sequences and 763 626 fungal sequences were obtained,clustering into a total of 5 284 bacterial OTUs and 709 fungal OTUs.The dominant bacterial phyla were Firmicutes and Proteobacteria,with Buchnera,Staphylococcus,and Acinetobacter as the predominant genera.As for fungi,Ascomycota and Basidiomycota were the dominant phyla,with Cyberlindnera as the most abundant genus.Comparative analysis revealed that in the bacterial community,winged fourth-instar nymphs exhibited significantly higher Firmicutes abundance,reduced Buchnera abundance,and increased Staphylococcus abundance compared to that of wingless nymphs,while no significant differences were observed between winged and wingless adults.In the fungal community,winged adults showed significantly lower Ascomycota and Cyberlindnera abundance than that of wingless adults,but no significant differences were found in fourth-instar nymphs.[Conclusion]These findings suggest that symbiotic microbiota may play a role in the wing dimorphism of S.miscanthi,providing a theoretical basis for elucidating the regulatory mechanisms of wing differentiation and developing novel symbiotic microbe-based pest control strategies.
