An epidemiological investigation and phylogenetic analysis of canine parvovirus in stray dogs around the Poyang Lake

Abstract
[Objective]This study aimed to understand the prevalence of canine parvovirus(CPV)and the genetic variation characteristics of CPV in stray dogs around the Poyang Lake.[Method]This study was conducted in January to June 2023 to collect a total number of 200 fecal samples from stray dogs in the Poyang Lake area.Colloidal gold immunochromatographic tests were used for the preliminary screening of antigens,from which 10 positive samples were selected for PCR amplification of the full-length CPV gene.The nucleotide sequences were determined,and bioinformatics software was used to analyze the amino acid sequences of the VP2 gene and construct a phylogenetic tree,followed by homology difference analysis and amino acid variation site analysis.[Result]Results showed that 56 positive fecal samples were identified through the colloidal gold antigen test,resulting in a positivity rate of 28%.Gene segment amplification yielded 10 full-length CPV sequences,named CPV-NC-2,CPV-NC-7,CPV-NC-8,CPV-NC-9,CPV-NC-17,CPV-NC-29,CPV-NC-31,CPV-NC-32,CPV-NC-34,and CPV-NC-35.Sequence analysis of the VP2 gene indicated that the amino acid sequence homology ranged from 98.4%to 99.7%.Multiple amino acid residue variations were observed among the strains.Phylogenetic analysis revealed that 8 strains belonged to the 2c subtype,while 2 strains belonged to the 2a subtype.[Conclusion]This study indicates that two subtypes of CPV are prevalent in the Poyang Lake area,and mutations have occurred.This study provides data support for the prevention and control of canine parvovirus in the vicinity of the Poyang Lake.关键词
