Abstract
Strain degeneration poses a significant challenge to the cultivation of edible fungi,however,the molecular mechanisms driving strain degeneration in these organisms remain relatively underexplored.In the present study,RNA interference technology was utilized to silence the long-chain acyl-CoA synthetase gene Le ACSL in Lentinula edodes.This approach successfully produced transformants exhibiting a 70%-90%reduction in gene expression.Examination of these transformants'growth on agar plates and within spawn bags revealed a 30%-40%decrease in growth rate,with colonies displaying varying degrees of sectoring and a stable phenotype.Notably,the transformants were unable to progress to the browning stage of mycelial film formation in spawn bags,leading to mycelial recession and inability to produce fruiting bodies.Microscopic analysis revealed increased branching and intertwining of the transformant mycelium,accompanied by severely wrinkled surfaces.Certain cells were observed to be entirely vacuolated and lysed,with organelles displaying compromised structural integrity.Subsequent analyses of fatty acid composition and malondialdehyde levels in the transformants revealed a 18%-29%reduction in the degree of fatty acid unsaturation and a 36%-54%decrease in lipid peroxidation compared to the wild type.These findings preliminarily suggest that the Le ACSL gene is critically involved in the strain degeneration of L.edodes.关键词
